摘要
目的初步研究一氧化氮(NO)供体硝普钠(SNP)对高胆固醇血症兔Oddi括约肌(SO)的作用及可能机制。方法新西兰大耳白兔24只,给予高胆固醇饮食饲喂8周后取SO组织制备成SO肌环,通过等长张力记录方法观察SO肌环初始收缩反应,并观察SNP对SO肌环的舒张作用;分离培养SO细胞,用HE及TUNEL染色观察SNP对SO细胞凋亡的影响。结果 (1)SO肌环初始收缩平均波幅为(0.144±0.004)g,平均收缩频率为(10.8±1.1)次/min;SNP对SO肌环有明显舒张作用,且具有浓度依赖性。(2)SNP处理后的SO细胞HE和TUNEL染色示明显的细胞凋亡征象。结论 NO供体SNP可能通过诱导高胆固醇血症兔SO细胞凋亡显著降低SO张力,其可能为SNP治疗Oddi括约肌功能障碍的机制之一。
Objective To investigate the effect and possible mechanism of nitric oxide(NO) donor sodium nitroprusside(SNP) on smooth muscle motility and cell apoptosis of sphincter of Oddi(SO) in the hypercholesterolemic(Ch) rabbits.Methods A total of 24 New Zealand rabbits were fed with high-cholesterol diet for 8 weeks.SO muscle rings were dissociated from the rabbits in vitro.Recording the contractile response and the relaxant effect by using SNP in the method of the isometric tension recording.Sphincter of Oddi cells were isolated and cultured.The effect of SNP on sphincter of Oddi apoptosis was detected by HE staining and TUNEL methods.Results(1) The initial contraction of the average volatility was(0.144 ± 0.004) g,the average contraction frequency was(10.8 ± 1.1) per minute.SNP had a concentration-dependent significant relaxant effect on SO muscle rin.(2) HE staining and TUNEL showed obviously apoptosis sign following SNP treatment.Conclusions The NO donor-SNP may relax the SO muscle tension through inducing SO cells apoptosis.
