摘要
目的观察诱发大鼠肝癌过程中PINCH1和F-actin蛋白表达特点,探讨其在肝癌发生发展过程中的作用。方法利用二乙基亚硝胺(DEN)制备诱发性肝癌动物模型,采用免疫组织化学方法检测PINCH1和F-actin蛋白在对照组、模型Ⅰ组(6周)、模型Ⅱ组(14周)、模型Ⅲ组(22周)大鼠肝脏中表达的差异。结果在对照组大鼠肝脏中少见PINCH1和F-actin蛋白表达阳性的细胞,其在模型Ⅰ组(6周)中主要表达在肝小叶周边的损伤区的周围。在模型Ⅱ组(14周)分布于门管区、肝小叶的周边及非典型增生结节内,其中PINCH1位于非典型增生结节内居多,而F-actin主要分布于基质细胞及细胞外基质中。在模型Ⅲ组(22周),PINCH1蛋白在肝癌结节周边区域呈过表达,癌周组织中也可见PINCH1蛋白表达阳性的细胞;F-actin主要表达在癌周的肝细胞、基质细胞细胞和细胞外基质中。模型组(22周)的PINCH1和F-actin蛋白表达的平均光密度值均高于其他各组。结论在诱发肝癌发生过程中PINCH1和F-actin蛋白表达异常,可能参与了肝癌发生发展过程。
Objective To investigate the role of PINCH1 and F-actin in the process of hepatocarcinogenesis according to PINCH1 and F-actin protein expression characteristics.Methods Induced liver cancer model was established by diethylnitrosamine(DEN).On this basis,the expression of PINCH1 and F-actin protein was observed with immunohistochemistry in the rat liver of control group,model groupⅠ(6 weeks),model groupⅡ(14 weeks) and model groupⅢ(22 weeks).Results The positive expression cells of PINCH1 and F-actin protein was rare in the liver of control rats.In the model group Ⅰ(6 weeks) they mainly distributed in the zone of liver damage around hepatic lobule.In the model groupⅡ(14 weeks),the positive cells of PINCH1 and F-actin protein distributed in the portal area,periphery of the hepatic lobule and atypical hyperplastic nodules.The PINCH1 located mostly within the atypical hyperplastic nodules,and F-actin mainly distributed in the stromal cells and extracellular matrix.In the model groupⅢ(22 weeks),the PINCH1 protein expression over expressed in liver nodules surrounding area,and protein-positive cells could also be found in the cancer tissues.And the cells of F-actin expression distributed in peritumoral liver cells,stromal cells and extracellular matrix.The average optical density of the PINCH1 and F-actin protein expression of the model groupⅢ(22 weeks) was higher than the other groups.Conclusions PINCH1 and Factin protein expression is abnormal in the process of induced liver cancer,which may be involved in the development of liver cancer.
