摘要
目的探讨凋亡抑制蛋白(IAP)家族的新成员Livin在非小细胞肺癌(NSCLC)中的表达及p53、bcl-2之间的关系。方法采用免疫组织化学法(SP法)检测45例NSCLC患者的肺癌组织,35例癌旁组织和15例肺良性瘤样病变的肺组织中Livin及p53、bcl-2的表达,使用χ2检验比较各组间差别;使用Spearman相关性分析分析Livin与p53,bcl-2之间的相关性。结果 Livin在45例NSCLC组织中62.22%表达阳性,而35例癌旁正常肺组织和15例肺良性瘤样病变组织低表达(分别为2.86%、6.67%,P<0.01)。Livin蛋白的表达与肺癌患者的病理类型、肿瘤分化程度、淋巴结转移及TNM分期均无明显相关关系(分别为P=0.844,P=0.156,P=0.565,P=0.537)。p53、bcl-2蛋白表达均与Livin蛋白表达呈正相关(p53:r=0.375,P=0.011;bcl-2:r=0.565,P<0.001)。结论 Livin在NSCLC组织中特异性的高表达提示其可能参与了NSCLC的发生,使其将来可能成为NSCLC诊断及治疗的新靶点。Livin与p53及bcl-2在NSCLC的发生中有可能发挥了协同作用。
Objective To examine and analyze the expression of Livin,a novel inhibitor of apoptosis protein(IAP) family member,in non-small cell lung cancer(NSCLC) and the correlation with p53 and bcl-2.Methods The expression levels of Livin and p53,bcl-2 in 45 NSCLC,35 paracancerous,15 benign tumor were detected by immunohistochemical staining(SP).Results Livin was expressed in 28 of 45 NSCLC(62.22%).Livin was expressed low levels in paracancerous or benign tumors(2.86%,P < 0.01 and 6.67%,P < 0.01).Livin expression had no significant relationship with clinically pathological characteristics of lung cancer patients.p53 and bcl-2 expression had a positive correlation with the expression of Livin(p53:r = 0.375,P = 0.011;bcl-2:r = 0.565,P < 0.001).Conclusions Livin may play a role in the carcinogenesis of NSCLC,which may serve as a novel target for NSCLC diagnosis and treatment.The overexpressions of Livin and p53,bcl-2 may play synergetic roles in process of carcinogenesis of NSCLC.
