不同来源阿萨希毛孢子菌菌丝相体外诱导条件的探讨 被引量:3

摘要 目的探讨在不同诱导因素条件下,不同来源的阿萨希毛孢子菌(Tricho sporon asahii,T.asahii)菌丝诱导结果的差异。方法 11株不同来源的T.asahii,选用RPMI-1640液体培养基、YPD液体培养基、吐温酵母肉汤(TYB)培养基和50%(v/v)胎牛血清,分别在15℃、25℃和37℃条件下培养,于1h、2h、3h、6h、12h、24h计数菌丝生成率。结果 T.asahii 15℃菌丝生成率均值为(0.15±0.42)%,25℃为(5.75±3.48)%,37℃为(33.81±15.39)%,差异有统计学意义(P值均<0.05)。RPMI-1640培养基诱导菌丝生成率均值为(46.24±25.50)%,YPD液体培养基为(36.28±21.85)%,TYB培养基为(33.93±21.29)%,50%(v/v)胎牛血清为(18.60±14.58)%,组间比较有显著统计学差异(F=6.29,P=0.0013),组内比较有显著统计学差异(F=21.80,P=0.0000)。随着诱导时间的延长,各菌株菌丝生成率也随之上升。结论综合比较各诱导因素,RPMI-1640培养基、37℃诱导24h可使大多数T.asahii菌株由孢子相获得较纯的菌丝相,而菌株来源的不同也是影响诱导结果差异的重要因素之一。 Objective To investigate the variance of hypha development in Trichosporonasahii (T.asahii),which from different sources and under different induction conditions.Methods 11 strains of T.asahii isolated from different sources were introduced.RPMI-1640 liquid media,YPD liquid media,tween yeast broth (TYB) medium and 50% (v/v) fetal calf serum were used to induce the T.asahii hypha development at 15℃,25℃ and 37℃.The hyphal formation rate was calculated at 1,2,3,6,12 and 24 hours respectively.Results The mean hyphal formation rate and standard deviation at different temperature was (0.15±0.42)% for 15℃,(5.75±3.48)% for 25℃ and (33.81±15.39)% for 37℃,the last one was significant higher than the other two (P<0.05).The mean hyphal formation rate and standard deviation in differernt medium was (46.24±25.50) % in RPMI-1640 medium,(36.28±21.85) % in YPD liquid medium,(33.93±21.29) % in TYB medium and (18.60±14.58) % in 50%(v/v) fetal calf serum.Significant difference was existed between every 2 groups (F=6.29,P=0.0013),and intragroup comparison also showed remarkable difference (F=21.80,P=0.0000).With the extension of the culture time,hyphal formation rate also rised in all T.asahii strains.Conclusion Incubation with RBMI1640 liquid media at 37℃ for 24 hours,most T.asahii strains could convert from pure spore phase to purer hypha phase easily.The source where the strain isolated from is another important factor which affect the hyphal induction.
