用红细胞吸附释放法纯化H9N2亚型禽流感病毒

Purification of H9 Subtype Avian Influenza Virus (AIV) RBC Method

制备50%的红细胞悬液,将H9N2禽流感病毒尿囊液(HA≤29)采用鸡红细胞吸附释放法进行纯化,然后分别将纯化的病毒液及弃去的上清进行血凝试验测定。结果表明,使用50%的鸡红细胞悬液基本能将病毒吸附完全,HA效价提高23倍。因此鸡红细胞吸附释放法是一种经济实用、操作简便,并有较高纯化效率的方法。

An absorption-release method with 50% red blood cells(RBC) was used to purify AIV(HA≤29)in allantoic fluid and the HA titers of precipitation and supermate were examined respectively.The result showed that the 50% RBC could effectively absorb almost all the virus with higher purity and HA titer increased 23 times.Therefore,the absorption-release method is a simple,economical and efficient way for small volume influenza virus concentration.
