铀与牛血清白蛋白相互作用研究

摘要 铀是长寿命核素,如果进入环境或人体会造成严重的危害。血清白蛋白是哺乳动物体内参与多种内源性和外源性物质的存储和转运的蛋白,具有重要的运载、缓冲和营养功能。为了探讨铀与血清蛋白在生理条件下的作用定量关系、影响因素与作用机理,本研究采用三因素四水平的正交表设计实验,发现醋酸双氧铀与牛血清白蛋白在pH3.0,醋酸双氧铀的浓度为0.60mg/mL,BSA的浓度为0.45mg/mL时作用最强。该研究还采用红外光谱分析了铀与牛血清白蛋白相互作用的可能机理。 Uranium is a long-lived radionuclide and can be an ecological and human health hazard if exposed into environment or body. Serum albumin is involved in the transportation and storage of both exogenous and endogenous substances,and also plays a significant role in carrying,buffering and nutrition. In order to study the interaction between uranium and serum albumin,as well as the in vivo mechanism of uranium metabolic pathway and biological effect,the orthogonal design experiments in four levels and the three factors were carried out. The results showed that the significant reaction conditions between uranyl acetate and BSA(Bovine Serum Albumin) was pH 3.0,when uranyl acetate concentration was 0.60 mg/mL,BSA concentration was 0.45 mg/mL. In this research the FTIR spectra were also used to analysis the interaction between the uranium and BSA.
