溶剂对卷烟烟气气相物捕集和细胞毒性检测的影响

Effects of solvents on gas phase trap of cigarette smoke and cytotoxicity assays

通过对不同溶剂和溶剂捕集液对卷烟烟气毒理学评价中常用细胞株的毒性作用研究,为卷烟烟气气相物捕集时选择高捕集率、低细胞毒性的溶剂提供依据.采用中性红细胞毒性法,研究了10种常用溶剂对中国仓鼠卵巢细胞的毒性作用,以及细胞毒性作用较小的溶剂的烟气捕集液对5种常用细胞株的毒性作用.在检测剂量范围内对中国仓鼠卵巢细胞的毒性作用结果为:磷酸缓冲液和乙酸乙酯无毒性作用;二甲基亚砜、70%甘油、无水乙醇、甲醇和丙酮的毒性作用较小;正丁醇、异丙醇和二氯甲烷的毒性作用较大.70%甘油、乙酸乙酯和丙酮的烟气气相物捕集液对5种细胞株的毒性作用大小不相同,其中70%甘油对卷烟烟气气相物的捕集效率相对较高,本身毒性较低,是较为理想的卷烟烟气气相物捕集溶剂.

The cytotoxicity of different solvents and cigarette smoke trapped by solvents to common cell lines in cigarette smoke toxicologic tests was studied to provide a basis for choosing solvents of high smoke trap efficiency and low cytotoxicity.Using the neutral red uptake assay,ten solvents were selected to study their cytotoxicity to Chinese hamster ovary cell line(CHO cell line) .Three solvents of low cytotoxicity were chosen to trap gas phase of cigarette smoke for studying the cytotoxicity in five common cell lines used in cigarette smoke toxicologic tests.The results of the neutral red uptake assays in CHO cell line indicated that PBS and ethyl acetate had no cytotoxicity;DMSO,70% glycerol,ethanol,methanol and actone had low cytotoxicity;butyl alcohol,isopropanol and dichloromethane had high cytotoxicity.The cytotoxicity of gas phase of cigarette smoke trapped by 70% glycerol,ethyl acetate and acetone,respectively,was different.Because the efficiency of 70% glycerol to trap the gas phase of cigarette smoke was relatively high and the solvent has low cytotoxicity,the solvent was suitable to trap the gas phase of cigarette smoke in cytotoxicityl assessment.
