常绿萱草组培快繁技术研究

Study on Tissue Culture and Rapid Propagation of Hemerocallis Hybrida

以常绿萱草茎尖为外植体,研究了不同消毒方式、培养基配方对于外植体生长的影响,并探索了最适于愈伤组织形成、不定芽增殖和生根的培养基配方。结果表明:在无菌条件下,用75%酒精消毒30S,再转入0.1%升汞溶液浸泡8分钟消毒效果最好;有利于外植体产生愈伤组织的培养基配方是MS+2.0mg/LBA+0.2mg/LNAA;适合愈伤组织诱导生芽培养基配方是MS+1.0mg/L2.4-D+1.0mg/LKT;适合幼苗生根的培养基配方是MS+NAA0.5mg/L+活性炭0.2mg/L。

The stem apex of Hemerocallis Hybrida was taken as explant cultured in vitro.The effects of different disinfection way and medium formula on explant growth Disinfection way was studied.The results showed that with 75% ethanol disinfection 30S,again into eight minutes HgCl2 solution soaking 0.1% disinfection was the best way;MS+2.0mg/L BA + 0.2mg/L NAA solid culture medium was advantageous to induce callus;after cultured in MS+1.0 mg/L 2.4-D + 1.0 mg/L KT solid culture medium was suitable for callus induction born bud;and MS+NAA0.5 mg/L+ Activated carbon 0.2 mg/L solid culture medium was the best medium to induce root.
