木聚糖降解酶酶法制取低聚木糖的初步研究 被引量:4

摘要 木聚糖酶是一类复合酶系,木聚糖水解酶具有多重性现象.酶解法的关键在于木聚糖酶对底物的适应性,即选择合适的木聚糖酶.着重介绍木聚糖的预处理、选择相关条件酶解.正交试验表明,木聚糖酶水解木聚糖的条件为:在摇床转速为220r/min,温度为45℃的条件下,50mL缓冲液(pH=3.6)中加入0.02%木聚糖酶酶解2g粗木聚糖5h,得到低聚木糖浓度为4.12mg/mL,低聚木糖占总糖浓度的41.18%. Xylan enzyme is a compound enzyme system.Xylan hydrolytic enzyme owns multiplicity.The key of zymohydrolysis is the adaptability of xylanase to substrate,that is,suitable xylanase should be chosen.Pretreatment of xylan and correlative condition selection of enzymolysis had been focused on in this paper.The orthogonal experiment showed that the optimum conditions of the xylan was enzymatic hydrolyzes into crude xylooligosaccharides with xylanases were at 45℃,pH=3.6,the agitation rate was 220r / min,the reactive time was 5 hours,xylan 2g/50mL and xylanase lording 2%(v/v).
