系统性红斑狼疮患者T淋巴细胞CD69的表达及意义

摘要 目的观察T淋巴细胞CD69抗原表达与系统性红斑狼疮(systemic lupus erythematosus,SLE)抗体的产生及疾病活动的相关性。方法利用流式细胞术,采用免疫荧光染色方法分别检测植物血凝素(PHA)刺激20h后30例SLE患者及15名健康对照者外周血CD4+、CD8+T淋巴细胞亚群CD69抗原的表达,同时检测SLE患者外周血抗dsDNA抗体、疾病活动度用SLEDAI记分。结果经PHA刺激后,SLE患者外周血CD4+、CD8+T淋巴细胞亚群CD69抗原的表达明显高于正常对照组(p<0.01),特别是活动期,并且CD69抗原的表达与SLE的活动性成正相关;30例患者中有22例抗dsDNA抗体阳性,抗体阳性组T淋巴CD69抗原的表达高于抗体阴性组(p<0.05)。结论活动期SLE患者T淋巴细胞CD69抗原的表达明显上调,CD69抗原的表达在T细胞活化过程中起重要作用,CD69抗原的表达与SLE的活动性呈正相关,可为判断狼疮的活动提供参考。 Objective To observe the rdativing between the expression of CD69 antigen on T lymphocyte and production of anti-dsDNA antibody and activity in systemic lupus erythematosus(SLE) ratients.Methods Nourished with phytohemagglutinin(PHA) for 20 hours,CD69 antigen on T lymphocyte subsets in the peripheral blood of 30 active SLE patients and 15 healthy individuals were determined by using immunofluoresence staining method and flow cytometry.The serum levels of anti-dsDNA antibody、were also determined.The activity of SLE was valued by SLEDAI.Results The expression of CD69 antigen on CD4+、CD8+T lymphocyte subsets of SLE patients was significantly higher than that of healthy group(p<0.01),especially in the active cases.Moreover,they showed correlate positively with SLEDAI.At the same time,compared with that on those patients with negative anti-dsDNA antibody,the expression of CD69 antigen on the patients with positive anti-dsDNA antibody was high(p<0.05).Conclusion These results suggest that the expression of CD69 antigen on T lymphocyte subsets of SLE patients ascends,especially in the active cases.the expression of CD69 antigen on T lymphocyte subsets play an important role in the activity of T-cell and correlates with activity of SLE.It may provid an potential indicator for activity evaluation in SLE patients.
