摘要
目的研究差异表达新基因BQ135232的结构功能以及真核表达。方法应用生物信息学方法分析BQ135232 mRNA基因序列,初步了解其基本性状,构建表达载体,转染真核细胞(CHO/dhfr^-)。结果BQ135232具备作为免疫原的基本条件,构建表达载体pcDNA3-BQ-ORF-GPI,成功转染CHO/dhfr^-。结论完成差异表达新基因BQ135232的真核表达.为后续的蛋白功能研究奠定基础。
Objective To study the architecture,function,and expression of differential expression novel gene BQ135232 obtained by SSH.Methods By using bioinformatic tools,we primarily understood the basic characters of the novel gene BQ135232,then constructed expressing vector,and transfected in eucaryot- ic cell.Results The bioinformatic analysis indicated that BQ135232 possessed the possibility to be an im- munogen,subsequently we constructed the expressing vector pcDNA3-BQ-ORF-GPI,transfected CHO/dhfr- successfully,and verified by RT-PCR.Conclusion Accomplished the expression of novel gone BQ135232 ORF in eucaryotic cell,laid the groundwork for future protein function research.
