姜黄素诱导人胃癌细胞SGC-7901凋亡的作用机制 被引量:3

The mechanisms of apoptosis induced by curcumin in gastric cancer cells

摘要 目的探讨姜黄素诱导人胃癌细胞SGC-7901凋亡的作用及其相关机制。方法以不同浓度的姜黄素作用于胃癌SGC-7901细胞,利用倒置相差显微镜观察细胞生长形态变化,通过MTT检测细胞生长抑制率,流式细胞术检测细胞凋亡,Western blot检测胃癌细胞中Fas及survivin的表达情况。结果姜黄素能显著抑制体外培养的SGC-7901细胞的生长并呈量-效和时-效关系,流式细胞术检测到亚二倍体凋亡峰,细胞凋亡率增加,Western blot结果提示经姜黄素作用后Fas表达率上升,survivin表达率下降。结论姜黄素能抑制胃癌细胞SGC-7901的生长并促进其凋亡,姜黄素可能通过上调Fas及下调survivin的表达而诱导凋亡。 Objective To study the mechanisms of Curcumin-induced apoptosis on human gastric cancer cell line SGC-7901.Methods SC,C-7901 cells were treated with various concentrations of Curcumin and the growth inhibition rates of it were accessed by MTT method.Apoptosis of gastric cancer cells were in- spected by flow cytometry.The expression of Fas and survivin in gastric cancer cells were evaluated by west- ern blot.Results Curcumin could effectively inhibit the growth of gastric cancer cells in dose-dependent and time-dependent manners,the sub-peak appeared and the apoptotic rate was increased.The expressions of Fas was higher in Western blot,meanwhile,the expressions of survivin was decreased.Conclusion Curcumin could significantly inhibit the growth and induce apoptosis of gastric cancer cells(SGC-7901),Curcumin could probably through up-regulating Fas and down-regulating surviving to induce apoptosis.
