新基因KIAA0372在淋巴瘤中的表达及其生物信息学分析

摘要 目的研究淋巴瘤和反应性增生淋巴结基因表达的差异,并采用生物信息学方法对一条淋巴瘤高表达基因 KIAA0372进行初步分析。方法分别将淋巴瘤淋巴结标本和反应性增生淋巴结标本的 mRNA 与表达谱芯片杂交,通过信号扫描、处理后获得两者的表达差异基因。并用生物信息学方法对一条无功能研究的新基因 KIAA0372进行初步分析,预测它的基因结构、染色体定位、编码蛋白质的理化性质、亚细胞定位、蛋白质功能域等信息。并对多物种中的相似性蛋白进行了系统进化分析。结果表达谱芯片共发现了152条差异表达基因。对淋巴瘤相关新基因 KIAA0372的上述性质进行了有效的预测,基本明确了该基因编码一胞外分泌蛋白,可能作为 TGFβ信号通路的一个配基参与淋巴瘤细胞的生成。结论表达谱芯片技术是一种研究肿瘤基因表达差异的有效的高通量研究方法。新基因 KIAA0372是一个有研究价值的新靶点。 Objective To explore the differentially expressed genes between lymphoma and reactive lymph node,and perform an initial bioinformatics analysis on a novel gene,KIAA0372,which is highly ex- pressed in lymphoma.Methods The mRNA of the tissues from the lymphoma and reactive lymph node were marked with biotin respectively and hybridized with expression profile mieroarray,and the differentially ex- pressed genes were obtained.Initial bioinformatics analysis was performed on a novel gene named KIAA0372. Its gene structure,genomic localization,the physical and chemical characteristics of the putative protein,sub- cellular localization,functional domain and so on were predicted,and the systematic evolution analysis was performed on the similar proteins among several species.Results By using the expression profile microarray, many differentially expressed genes were uncovered.The efficient bioinformatics analysis was fundamentally i- dentified that KIAA0372 is a cytosol protein,and may involved in the protein degrading as metallopeptidase. Conclusion Mieroarray is an efficient and high throughout strategy for the detection of differential expressed genes.KIAA0372 is considered as a potential target for tumor metastasis researches by bioinformatics analysis.
