摘要
目的研究人骨髓基质干细胞经体外培养后细胞的表型。方法8例成年志愿者(无系统性疾病,20~40岁)髂前上嵴骨穿法得人骨髓,密度梯度法分离骨髓基质干细胞(human bone marrow mesenchymal stem cell,hMSC),单纯DMEM培养液,分别培养至第2代细胞,免疫荧光和流式细胞仪鉴定hMSCs细胞表型,CD105,CD166,CD29的表达。结果第二代细胞可见明显CD105,CD166,CD29表达,第二代细胞CD105阳性率78±6%,CD166阳性率43±7%,CD29阳性率69±12%。结论人MSC体外经传代培养后可表达干细胞特征表型。
Objective To study the cell surface antigens of human bone marrow mesenchymal stem cell (hMSC). Methods Human MSC were isolate from bone marrow, which aspirated from iliac crest and cultured in defined medium in vitro. hMSC at passage 2 were harvested and cell surface marker CD29、CD105、CD166 were assessed using FACS. Results 78±6% of the second generation cells showed positivity to CD105, The percentage of CD166 were 43±7% and 69±12% as CD29. Conclusion The second generation of hMSC cells can be used for the study of cells induced and differentiation and the seed-cells of clinical applications.
