摘要
目的研究用于组织修复的壳聚糖-明胶支架材料的制备及其性能,为其进一步的细胞培养以及组织工程皮肤的临床应用积累数据。方法将壳聚糖与明胶在一定条件下共混,预冻,冷冻干燥并交联得到支架材料,考察其性能。结果制备过程中,预冻溶液的固含量、壳聚糖与明胶的配比以及预冻温度等参数对支架材料的微观结构、表观密度、含水量和力学性能有较大影响。结论采用冷冻干燥法可以成功地制备壳聚糖-明胶支架材料,通过控制其反应条件可以调整支架孔隙结构和性能,从而得到满足需要的支架材料。
Objective To study the preparation and properties of chitosan-gelatin scaffolds used for tissue regeneration and provide some data for the cell culture and clinical application of tissue-engineered skin. Methods Chitosan and gelatin solutions were mixed together under certain condition, and then frozen, lyophilized, cross-linked to get scaffolds. The microstructure was observed by scanning electron microscopy (SEM) and the properties were characterized by apparent density, water uptake ability and tensile strength. Results It was found that the concentration of the solution, the ratio of chitosan to gelatin, and pre-freezing temperature have great effect on the microstructure, apparent density, water uptake ability and mechanical properties of the scaffolds. Conclusion Freezing-drying technique is an efficient way to prepare chitosan-gelatin scaffolds. The pore structure and the properties of the scaffolds can be adjusted by controlling the reaction conditions in order to satisfy various needs.
