羊膜抑制兔角膜眼表纤维化的研究 被引量:1

摘要 目的探讨羊膜移植抑制眼表纤维组织增生的机制。 方法将传代的兔角膜成纤维细胞接种于羊膜基质面 ,分别培养 1d和 7d ,采用吖啶橙法观察羊膜对成纤维细胞凋亡的影响。同时采用原位杂交法 ,观察成纤维细胞中bFGFmRNA表达的变化。 结果羊膜对培养 1d和 7d的兔角膜成纤维细胞均有明显地抑制凋亡作用。在羊膜基质面培养 1d的兔角膜成纤维细胞 ,其bFGFmRNA的表达与对照组相比无明显改变 ,而培养 7d后 ,bFGFmRNA的表达明显下降。 结论羊膜不仅有抑制体外培养的兔角膜成纤维细胞凋亡的作用 ,而且还能抑制体外培养的兔角膜成纤维细胞bFGFmRNA的表达 。 Objective To investigate the mechanism of the inhibition of ocular fibrosis by amniotic membrane transplantation. Methods The rabbit corneal fibroblasts after primary culture were inoculated on the amniotic membrane matrix for 1d or 7d. Fibroblasts apoptosis was studied by acridine orange. bFGFmRNA in the rabbit corneal fibroblasts were examined through hybridization in situ . Results The a mniotic membrane inhibited the apopotosis of fibroblasts significantly which has been on the amniotic membrane matrix for 1d or 7d. bFGFmRNA showed no change, but it was suppressed significantly when the fibroblasts were cultured on the amniotic membrane for 7d. Conclusion Fibro blasts apoptosis can be inhibited, and bFGFmRNA in the rabbit corneal fibroblasts in vitro can be suppressed by amniotic membrane matrix. This can partially explain the effect of the amniotic membrane on inhibition of fibrosis.
