摘要
目的探讨Ki-ras基因突变在肺癌不同病理型中的分布及与吸烟的关系。方法采用聚合酶链反应-单链构象多态性分析(PCR-SSCP)检测40例肺癌病理标本Ki-ras基因第12位密码子点突变。结果检出6例突变者,突变率为15%。腺癌和鳞癌的突变率分别为17.4%(4/23)和13.3%(2/15),无显著性差异(χ2=1.43,P>0.05)。有吸烟史的肺癌突变率为6.45%(2/31),非吸烟肺癌中突变率为44.4%(4/9)。结论在非小细胞肺癌中未发现Ki-ras基因突变与吸烟有关。
Objective To study the relationship between smoking related lung cancer and mutation in codon
12 of Kiras in nonsmallcell lung cancer. Method Single strand conformation polymorphism
analysis of polymerase chain reaction products (PCRSSCP) with silver staining was used to
detect the mutation in codon 12 of Kiras in the paraffinembedded lung cancer tissue. Results
Six of 40 cases (15 percent) were found with mutations in codon 12 of Kiras. The frequence of
mutation was 17.4 percent for adenocarcinoma and 13.3 percent for squamous cell carcinoma in
subtype analysis, respectively (P>0.05). The mutation was more frequent in nonsmokers
(44.4%) than in smokers (6.45%). Conclusion The results failed to prove that mutation in codon
12 of Kiras mutations is associated with tobacco smoking in nonsmallcell lung cancer.
