摘要
建立了染色混合精斑涂片DNA的提取方法,通过两步扩增的技术与垂直聚丙烯酰胺凝胶电泳及银染色法,成功检测了3个STR位点.结果显示,所有染色涂片DNA分型均与对照血样一致,苏木素-伊红(HE)染色比酸性复红-美蓝(Baecchi)染色对DNA的降解作用要小;涂片灵敏度检验可达到少于10个精子,且室温保存一年以内的涂片其灵敏度光显著差异.该方法简单、快速、可靠,为染色混合精斑涂片的PCR分析提供了新方法.
A method of extraction of DNA from stained smears of mixed seminal and vaginal stain was established by using the two - step amplification, polyacrylamide gel vertical electrophoresis and silver staining techniques. Three STR loci have been performed successfully. The results show that all DNA - typing of the stained smears were consistent with the corresponding blood; the smear slides stained with hematoxylin-eosic(HE) made the samples' DNA less degraded than stained with the acid fuchsin and methylene blue(Baecchi); a minimum of spermatozoa was less than 10 for positive results and the sensitivity of smears slides kept within 1 year at room temperature were no difference. The method is rapidity, simplicity and reliability for testing stained smears of mixed seminal stain.
