白细胞介素2(IL-2)基因和γ干扰素(IFN-γ)基因体内转染巨噬细胞对恢复化疗所致免疫功能损伤的研究

摘要 用磷酸钙——DNA共沉淀法, 将pREP8IFNγ真核表达质粒或/和ρREP8IL2 真核表达质粒直接注射至经大剂量化疗后的小鼠腹腔内, 成功地转染腹腔巨噬细胞(Mφ)。IL2 基因和IFNγ基因联合转染Mφ较IL2 或IFNγ基因转染Mφ, 其基因表达产物水平更高, 显著地刺激脾脏增生, 增强淋巴细胞的增殖, 增强腹腔Mφ的细胞毒活性, 促使腹腔Mφ分泌白细胞介素1 (IL1)、肿瘤坏死因子(TNF) 和NO。结果表明, 双基因转染效果优于单基因转染, IL2 和IFNγ基因联合转染Mφ更有助于由放疗、化疗所致受损的免疫功能恢复。 Using calcium phosphate coprecipitation method the eukaryotic expression vector p REP 8 IL 2 or/and p REP 8 IFN γ were peritoneally injected into mice after high dose chemotherapy, IL 2 gene or/and IFN γ gene were successfully transfected into peritoneal MΦs. The level of expression products of IL 2 gene or/and IFN γ gene in Mφs was higher than before. The gene expression products could incscase the proliferation of splenocytes, elevate the cytotoxicity of Mφs, and promote the secretion of IL 1, NO and TNF of Mφs. The results showed that IL 2 gene transfection in combination with IFN γ gene transfection was more potent in the enhancement of immune function than each of them transfected separately. This suggested that the combination of IL 2 and IFN γ gcne transfection into mφs might be helpful for the recovery of immune injury caused by chemotherapy or radiotherapy.\;
