摘要
本实验采用窦房结细胞纯化培养、免疫细胞化学、免疫组化、免疫电镜 (pAg 技术) 和图像分析等多种方法,研究了新生SD大鼠窦房结的ANF表达。结果提示: 窦房结原位组织和培养细胞的胞浆内均可见ANF阳性反应颗粒。培养细胞的核附近电子致密颗粒有10nm ANF免疫反应的pAg 颗粒。窦房结ANF的含量和活性明显低于心房 (P< 001)。
Using prue culture of cells of SAN, immunohistochemical and immunocytochemical reaction, immunoelectron microscope (pAg technique) and image analysis methods, the ANF expression in SAN cells of neonatal SD rats was studied. The results showed that ANF positive granules were in cytoplasm of tissue and cultured cells of SAN, 10 nm ANF immunoreactive pAg granules were in the electron dense granules near nuclei of cultured cells of SAN. The volume and activities of ANF in SAN were significantly lower than those in atrium (P<0 01). The physiological roles of ANF in SAN cells were also discussed.\;
