对大肠杆菌表达的重组人白细胞介素 4进行了分离纯化。人 IL- 4基因表达产物在大肠杆菌中以不溶性包含体形式存在 ,经过超声破菌、包含体抽提、洗涤处理 ,可使包含体纯度达 70 %以上 ,用 6mol/L盐酸胍变性溶解包含体沉淀后上清直接稀释复性 ,再经浓缩、透析、离子交换色谱一系列纯化步骤终产物纯度达 95 %以上 ,回收率 1 0 %以上 ,比活性达 2 .5× 1 0
rhIL 4 expressed as insoluable including bodies in E.coli by pBV220/hIL 4 were isolated and purified. After treatment of supersonic, inclusion bodies extraction and washing the purity of including bodies may be more than 70%. After denaturing and dissolving including bodies with 6mol/L guanide hydrochloride and precipitation, the supernate is diluted to get renaturized. After a set of purification steps of enrichment, dialysis and iron exchange chromatography, purity of the final product is over 95%, the recovery rate is over 10% , the ratio activity reach 2.5×10 6 U/mg protein.
