四物汤等复方多糖组分对小鼠腹腔巨噬细胞释放细胞因子的影响

Effect of polysaccharide ingredients from TCM complex prescription on releasing cytokines of mouse peritoneal macrophages

目的:观察四物汤等6个方剂多糖组分对小鼠腹腔巨噬细胞(Mφ)释放细胞因子(CK)水平的影响,探索上述复方疗效的可能机制,并为其活性成分的提取和筛选提供导向。方法:制备四物汤、四君子汤、六味地黄汤、玉屏风散、桂枝汤、龙胆泻肝汤等6个复方的总多糖组分,96孔板中检测其对小鼠腹腔Mφ释放白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、白细胞介素8(IL-8)、肿瘤坏死因子α(TNF-α)等细胞因子(CK)的影响。结果:各复方多糖组分能明显促进小鼠腹腔Mφ释放上述一个或多个CK,其效应具有浓度差异性(P<0.05或P<0.01);其中,相对于正常Mφ,复方多糖能促进经LPS诱导的Mφ释放更多种类的细胞因子。结论:6个复方多糖组分能促进小鼠腹腔Mφ释放不同的CK,可能与上述方剂的疗效机制有关,这为分离其活性成分提供了导向;同时也为建立中药活性成分的Mφ筛选模型提供了依据。

Objective： To observe the effect of polysaccharide ingredients from six TCM complex prescriptions on releasing the cytokine（CK） level of mouse celiac macrophages （Mφ）, to explore the possible mechanism and direct the extractionand separation of active components for above compounds. Methods： The total polysaccharide ingredients of six complex prescriptions were prepared： Siwu Decoction, Sijunzi Decoction, Liuweidihuang Decoction, Guizhi Decoction, Longdanxiegan Decoction and Yupingfeng Pulveres, then the mouse celiac Mφ were incubated together in 96 holes board. Furthermore, levels for the latter to release CK, including IL-1？, IL-6, IL-8 and TNF-α, were measured. Results： Polysaccharide ingredients of each complex prescription could obviously accelerate mouse celiac Mφ to release one or more CK, and the e ects had concentration otherness （P〈0.05 or P〈0.01）. The polysaccharide complex could accelerate Mφ induced by LPS to release more sorts of CK than the normal Mφ. Conclusion： All polysaccharide ingredients of six complex prescriptions could accelerate the mouse celiac Mφ to release di erent CK. It was probably related with the treated mechanism of six complex prescriptions. This could direct the extraction and separation of active components and could provide with a gist for establishing screening model of Mφ for active component.
