中间普氏菌内毒素脂多糖的细胞免疫介导活性

Cellular Immunological Activity of Endotoxin Lipopolysa charide Produced by Prevotella Intermedius

采用酚水法提取中间普氏菌ＡＴＣＣ２５２６１内毒素脂多糖，通过Ｋｒａｍｅｒ测定法，软琼脂细胞培养法、胸腺细胞增殖法以及局部斯氏反应测定脂多糖的免疫细胞生物学活性。结果显示，纯化脂多糖可显著地诱导小鼠模型生成肿瘤坏死因子（ＴＮＦ）、集落刺激因子（ＣＳＦ）和白介素－１（ＩＬ－１），并在一定范围内呈剂量依赖型。同时中间普氏菌内毒素还可刺激皮肤形成典型的局部斯氏反应。

おellular immunological activities of endotoxin prepared from Prevotella.Intermedius (P i)ATCC 25261 were examined in the production of interleukin 1 (IL 1),tumor necrosis factor (TNF),colony stimulating factor(CSF) and Shwartzman preparatory Dose50(SPD50).The results demonstrated that IL 1 produced at dose 0.1ng of endotoxin,and the level of IL 1 was correlated with the dose of endotoxin.The endotoxin also induced the production of sera TNF at a dose of 10μg in mice.TNF was found in sera of mice after the endotoxin injection one hour later,and disappeared within six hours.The results also show that this endotoxin induced mouse macrophages to produce CSF at the dose 0.1μg and have a dose dependent response,the SPD50 of Pi endotoxin was 64μg in local Shwartzman reaction.Those results demonstrated that the activities of Pi endotoxin on cytokins may be important to its pathogenesis in clinical infection.
