摘要
实验选择黄嘌呤──黄嘌呤氧化酶作为系统,反应在O2存在的条件下,废物黄嘌呤发生催化氧化,产生超氧自由基(O2.-),由2.6-二氯靛酚钠试剂检测。通过运用Michaelis反应机理处理,最终得到米氏常数(KM)。Cu2+能抑制黄嘌呤氧化酶的作用效果,其抑制作用可通过v0-[Cu2+]曲线看出。
Xanthine-xanthine oxidase is chosen as system for this experment. The reaction is a catalytical exidation of the supstiate xanthine in the presence of O2, the generated supevoxide raclical (O2.-) is examined by the 2, 6 -Dichloroindophenol sodium. The Michaclis constant then can be finally concluded through mechanism treatment of Michaelis reaction. Cu2+ can inhibit the effeet of xanthine oxidase. The inhibition can be obseived from v0[Cu2+] curve.
