银杏提取物对心肌细胞缺氧缺糖和氧糖复供时钙内流的抑制作用

摘要 目的:观察银杏提取物(GbE)对心肌细胞缺血和缺血/再灌注损伤时细胞钙内流的影响。方法:以心肌细胞缺氧缺糖模拟缺血,用缺氧缺糖后一定时间氧糖复供作为再灌注,采用改良“冷镧”法测定细胞^(45)Ca 摄取率。结果:GbE 在一定浓度(35-350mg/L)范围内可显著降低缺血和缺血/再灌注时细胞^(45)Ca 摄取,其抑制作用呈一定的量效关系。结论:GbE可能具有通过抑制钙内流而减轻缺血和缺血/再灌注后心肌细胞损伤的作用。 Objective:To investigate the effect of Ginkgo biloba extract(GBE)upon calcium influx in eardiomyoeytes injured by ischemia and reperfusion.Methods:The cells were cultured in depletion of oxygen and glucose and resupply of them which simulated ischemic and reperfusional injury.Cellular ^(45)Ca-uptake was measured with the modified Lanthanum method.Results:GBE(35~350 mg/L)could inhibit cellular ^(45)-uptake in the cultured cardiomyocytes in the course of the simulated ischemic and reperfusional treatments significantly in dose-dependent fashion.Conclusion:GBE might play a role in myocardial ischemic and reperfusional injury through its inhibitory effect on calcium influx.
