红芪多糖对培养血管壁平滑肌细胞的影响

摘要 目的:观察红芪多糖对培养血管壁平滑肌细胞(VSMC)的影响。方法:应用组织贴块法,在正常培养条件和高脂血清损伤等条件下,观察红芪多糖对 VSMC 的增殖、自由基清除及超微结构等的影响。结果:在正常、高脂血清、巨噬细胞孵育液培养条件下,有显著抑制 VSMC 的作用(P<0.01);对肾系膜细胞孵育液、未融合状人内皮细胞孵育液及 VSMC 孵育液培养条件下刺激的 VSMC 增殖无显著抑制作用(P>0.05);对正常、高脂血清培养条件下的VSMC 内过氧化脂质可显著降低(P<0.01),对超氧化物歧化酶活性可以显著增高(P<0.01),并能提高高脂血清培养条件下前列环素/血栓素 B_2比值。结论:红芪多糖可在高脂血清培养条件下,保护 VSMC 形态结构,促进细胞新陈代谢,有益于延缓动脉粥样硬化的进程。 Objective:To observe the inhibiting effect of Hedysarum polybotrys polysaccharose(HPP)on the proliferation of vessel smooth muscle cell(VSMC)in different cultivating conditions:In normal serum,hyperlipidic serum(HLS),and in macrophage-incubating fluid.Methods:Using trypsase digestive ferment to cultivate VSMC of human or rabbits in vit- ro for observation.Results:In cultivating conditions of mesangium cell-incubation fluid,umbilical cord vessel endothelial cell-incubation fluid and VSMC incubation fluid,HPP had no evident inhibiting effect on proliferation of VSMC.It could evidently reduce lipid peroxide in VSMC and increase the activity of superoxide dismutase in the cultivating conditions of normal serum and 3% HLS serum.In HIS cultivating condition,lAPP could increase the ratio of PGI/TXB so that it could prevent and cure arteriosclerosis.Conclusions:In the cultivating condition of hyperlipidic serum,HPP could protect the morpbosis of VSMC,promote the metabolism of cells,which contributes to the delaying of arteriosclerosis.
