抗纤方抗肝纤维化模型大鼠肝细胞损伤的实验研究

摘要 目的:探讨抗纤方阻断肝纤维化的机理。方法:从肝纤维化模型大鼠的肝组织中分离肝细胞,用抗纤方培养,并与秋水仙碱、大黄虫丸培养组进行对照观察。结果:抗纤方培养组肝细胞存活率明显高于空模组(P<0.01);抗纤方、秋水仙碱、大黄虫丸组细胞培养上清液中谷丙转氨酶的值均低于空模组(P<0.01);抗纤方、大黄虫丸组肝细胞中糖元、酸性磷酸酶含量明显高于空模组(P<0.01),而秋水仙碱则与空模组相当(P>0.05)。结论:抗纤方能保持细胞膜稳定,促进物质代谢,加快肝细胞修复再生,间接抑制胶原纤维合成,从而达到抗肝纤维化的作用。 Objective:To study the mechanism of the inhibiting effect of Kangxian recipe (KXR) on hepatic fibrosis.Methods: The hepatocytes isolated from the liver tissues of the rats with hepatic fibrosis were cultured with KXR,colchicine and Dahuang Zhechong Pill (DHZCP) separately.The KXR group was the observation group,and the colchicine and DHZCP group were the control groups.Results:The survival rote of hepatocytes of KXR group was higher than that of model group (P<0.01).The activities of alanine transaminase (ALT) in the supernatant in KXR group,colchicine group or DHZCP group were lower than that in the model group (P<0.01).The levels of glycogen and ACP level in KXR group or DHZCP group were higher than those in the model group (P<0.01),but in colchicine group these parameters was e- qual to those in the model group (P>0.05).Conclusion:KXR could keep the hepatocytes membrane steady,and pro- mote the metabolism of substances,and accelerate the repair of the hepatocytes and restrain indirectly the synthesis of col- lagen,and then prevent the liver from fibrosis.
