摘要
目的:了解红毛五加多糖促进造血细胞生成的作用机理。方法:选用脐带血单个核细胞,加入适量红毛五加多糖,观察长期液体培养系统中粒、单系祖细胞集落刺激因子(GM-CSF)的活性。结果:依量效曲线浓度为据,向培养体系中加入加脐血基质细胞上清液,对照组粒单祖细胞(GM-CFU,2×10~5/ml)为58.00±2.65,5%红毛五加多糖—脐血基质细胞上清液(hmwjdt-pscs),GM-CFU 为91.67±4.04,10%hmwjdt-pscs,GM-CFU 为111.00±11.79;两实验组与对照组比较呈现显著性差异(P【0.001)。为了鉴别红毛五加多糖直接刺激粒系祖细胞形成作用的影响,本研究还设了与作用基质细胞相同浓度的红毛五加多糖(10μg/ml),加于粒系祖细胞培养体系中以资对照。结果表明:红毛五加多糖没有直接刺激粒系祖细胞形成的作用。结论:红毛五加对粒系造血祖细胞促进形成作用,是通过刺激基质细胞分泌 GM-CSF 实现的。
Objective:To study the mechanism of Acanthopanax Giraldii polysaccharide(AGP)on formation of hematopoietic cells.Methods:AGP was added to single nuclear cells from human umbilical cord blood and the activity of granulo- macrophage colony-stimulating factor(GM-CSF)in long-term culture system was observed.Results:The supernatant of umbilical blood stromal cells(SUBSC)was added to culture system according to concentration-effect curves,GM-CFU(2 ×10~5/ml)in the control group was 58±2.65.In 5%AGP-SUBSC,it was 1.67±4.04,in 10%AGP-SUBSC,it was 111±11.79.There was significant difference between the experimental group and the control group,P<0.001.A con- trol group with 10 μg/ml AGP was used to study the direct effects of AGP on formation of granulo-macrophage-progenitor cells,and the results was negative.Conclusion:AGP could promote the formation of granulo-macrophage-progenitor cells through stimulating GM-CSF secretion.
