摘要
目的:为了解黄芪、白术及其组成的复方制剂壮骨肾宝对大剂量激素所致的类固醇性骨质疏松的作用。方法:采用氢化可的松3.6mg/kg,给大鼠灌胃给药,每周6次,1个月后改为每周2次,建立类固醇性骨质疏松动物模型。分别取黄芪、白术水提液各5g/kg,黄芪、白术的复方制剂壮骨肾宝1.65g/kg 灌胃给药,每周6次,持续60天,用于上述动物模型。用不脱钙骨制片,用图像定量法对大鼠胫骨近端骨小梁骨代谢的静、动态指标进行测算。结果:氢化可的松组骨吸收增加261%,骨形成率减少63%,骨小梁面积减少23%,出现明显的类固醇性骨质疏松。结论:黄芪、白术及其组成的复方制剂壮骨肾宝对这种大剂量激素所致的类固醇性骨质疏松均有显著的防治作用。
Objective:To study the preventive and curative effects of Radix Astragali (RA) ,Rhizoma Atractylodis Macro- cephalae (RAM) and Zhuanggu Shenbao (ZGSB,a complex prescription made of above mentioned 2 herbs) on glucocorti- coid-indueed osteoporosis.Methods:A group of 3-month-old male SD rats were given (gastrogavage) hydrocortisone 3.6 mg/kg,3 times per week for 30 days,then twice per week for 30 days so as to set up animal models with glucocorticoid- induced osteoporosis.The rats were treated with RA 5g/kg,or RAM 5g/kg,or ZGSB 1.66g/kg 6 times per week by gastrogavage.After 60 days,all rats were killed,undecalcified bone slices of the proximal tibiae of rats were prepared for histomorphometic analysis.Results:(1) In comparison with control rats,the bone resorption was enhanced (+261%), bone formation decreased(-63%),and trabecular bone areas were significantly reduced(-23% ) in rats who received hy- drocortisone.(2) Trabecular bone formation of rats treated with RA,RAM or ZGSB was significantly increased,the bone resorption was reduced,trabecular bone areas were increased when compared with those of rats who received hydrocorti- son.Conclusion:RA,RAM and ZGSB could prevent glucocorticoid-induced osteoporosis.
