摘要
本实验利用山羊睾丸间质细胞体外培养的方法,研究IGF-I对cAMP和睾酮的影响。结果表明:(1)IGF-I可使间质细胞外cAMP积聚能力显著增强( P<0. 05~ P <0. 005)。(2)IGF-I可使间质细胞分泌睾酮的能力显著增加(P<0.05~P<0.01)。(3)IGF-I可刺激山羊间质细胞经 hCG诱导睾酮分泌显著增多( P<0. 05~ P<0. 01)。以上结果与催乳素和白细胞介素-1α这些含氮激素(因子)调节睾酮分泌的机理不同。
Leydig cells from 20 to 60 days'goats were cultured in 24-well tissue culture plates in Medium 199 with or without hCG(0. 1 -3. 0 IU/mL)and/or IGF- I (1- 100 ng/mL). The culture media were changed every 24 or 48 h and saved at - 20℃ until assayed for testosterone and extracellular cAMP. The results showed that IGF- I promoted extracellular cAMP accumulation (P <0. 05 - P <0. 005), enhanced both basal and hCG-induced testosterone secretion(P <0. 05- P <0. 01 ). The effects of IGF- I were in dosedependent manner. It was different from the mechanism of prolactin (PRL) and interleukin-1α (IL-1α) regulation of testosterone secretion.
