摘要
目的:探讨甘油三酯和极低密度脂蛋白对大鼠肝星状细胞增殖的影响。方法:用链霉蛋白酶和胶原酶原位灌流,Nycodenz密度梯度离心分离大鼠肝星状细胞,并以MTT比色法观察甘油三酯和极低密度脂蛋白对肝星状细胞增殖效应。结果:甘油三酯和极低密度脂蛋白可影响肝星状细胞的增殖,甘油三酯和极低密度脂蛋白浓度分别在12.5μg/ml和25~100μg/ml时对肝星状细胞增殖有促进作用(P<0.05或P<0.01)。结论:体外细胞培养表明,甘油三酯和极低密度脂蛋白可促进肝星状细胞增殖,其与脂肪肝纤维化的发生可能有关。
Background/Aims: To study the effect of triglyceride (TG) and very low-density lipoprotein (VLDL) on proliferation of hepatic stellate cells (HSC). Methods: HSCs were isolated and cultured from liver of Wistar rats by in situ perfusion with pronase and collagenase and density gradient centrifugation with Nycodenz. MTT colorimetric assay was detected for HSC proliferation. Results: TG and VLDL had an effect on proliferation of HSC and in concentration of 12.5ug/ml TG and 25-100 ug/ml VLDL promoted HSC proliferation (P<0.05 or P<0.01). Conclusions: TG and VLDL may promote HSC proliferation and may be associated with fatty liver and hepatic fibrogenesis.
