本研究用逆转录/聚合酶链式反应(RT/PCR)技术检测脑型和诱导型一氧化氮合成酶(NOS)mRNA在Sprague—Dawley大鼠大脑皮质、垂体、小脑、心脏、肺脏、肝脏、脾脏、小肠、肾脏和肾上腺的表达。结果发现,脑型NOSmRNA表达于小脑、心脏和肾上腺组织;而诱导型NOSmRNA表达于中枢神经系统以外的所有受检组织。本文初步探讨了各组织NOSMRNA表达阳性的生理意义。
Mammalian cells are endowed with at least the genes encoding distict isoforms of nitric oxidesynthase (NOS):constitutive brain-type NOS, constitutive endothelium-type NOS, and inducible NOS. To determinethe expressions of brain-type and inducible NOS mRNAs in vivo, 10 organs from Sprague -Dawley rats weredetected by reverse transcription /polymerase chain reaction (RT/PCR ):cerebar cortes, pituitary , cerebelIum, heart.lung, liver, spleen, small intestine, kidney, and adrenal gland. The findings are that the brain-type NOS mRNAsare detected in cerebellum, heart, adrenal gland, and that the inducible NOS mRNAS can virtually be detected in alltissues observed apart from those of the central nervous system, i. e. cerebral cortex, pituitary, cerebellum. Theresult indicates that the expressions of brain-type and inducible NOS mRNA are different in rats. The possiblephysiologic roles for positive expressions of NOS mRNAs in various tissues are also discussed.
