摘要
本实验采用大鼠49只,常温肝缺血再灌注模型,对肝缺血不同时间及再灌注后肝组织化学活性变化进行了分组定性对比观察.结果显示:1.缺血期,肝细胞PAS反应和SDH、AKP.Mg^(++)-ATPase活性随缺血时间延长逐渐减弱,LDH活性于缺血初期有所增强,随后逐渐减弱,到缺血90min时,上述各酶组织化学活性及糖原(PAS反应)均几乎消失,而ACP活性则显著增强.2.再灌注期,肝酶组织化学活性及PAS反应的变化趋势与缺血时间长短有密切关系,缺血20、40min再灌注后,肝酶组织化学活性及糖原(PAS反应)可逐渐恢复正常水平,而缺血60、90min再灌注后,肝脏SDH、AKP、LDH、Mg^(++)-ATPase和PAS反应均消失,ACP活注进一步增强.
The present experiment adopted the model of normothermic hepatic blood flow clamping and declamping in rat. The enzymhistochemical changes in hepatocyte during ischemia and reperfusion were observed comparatively. The results showed that; 1. the reaction of PAS, SDH, AKP and Mg++-ATPase changed from strong to weak with prolongation of ischemia time. The activity of LDH was enhanced in the initial stage of ischemia, then it became weakened; up to 90 min. after ischemia, all the above enzymatic reactions were obsent, while ACP activity was stregthened significantly. 2. During reperfusion, the tendency of enzymhistochemical changes had close relation to the time of ischemia. When reperfusion was given after being ischemic for 20 and 40 minutes, The changes and PAS reaction could return to normal gradually, but if ischemia was prolonged to 60 and 90 minutes, ACP activicty was further enhanced and all the enzymatic reactions would disapper.
