摘要
用细胞培养、酶细胞化学、同位素掺入等技术对新型单克隆抗体(Anti-LA18)抗K562-红白血病细胞株的作用进行了观察.结果用含不同浓度Anti-LA18抗体的培养液培养24小时后,实验组K562细胞出现体积小,核质比缩小,核染色质浓缩、胞质内空泡增多等形态学改变.酶细胞化学方法显示实验组K562细胞酸性磷酸酶、氯醋酸AS-D酯酶、酸性醋酸酯酶等反应均有所减弱,以酸性磷酸酶反应减弱最为明显.以K562细胞各种酶阳性百分率表示酶活性变化指标,对照组与实验组具有明显差异(P【0.001).抑制胸腺嘧啶核苷结合法测定Anti-LA18抗体对K562细胞增殖抑制作用,呈现纯Anti-LA18抗体浓度的依赖性.18小时~3H-TdR释放法测定Anti-LA18抗体对K562细胞的细胞毒作用,显示出杀伤活性随Anti-LA18抗体浓度的增加而增加.
We have investigated the effects of anti-LAl8 on K562 cells using cell culture, enzyme cytochemistry and other techniques. The results showed that the number and volume of K562 cells in experimental group markedly decreased. The activities of acid phosphatase (ACPase), a-naphthyl acetic esterase (ANAE) and AS-D chlorides acetic esterase (AS-D) became weaker in K562 cells which were treated by different concentration of anti-LA18 antibody when compared with control group cells. The results of the groups were significantly different (P<0. 001). The method of 3H-TdR incorporation showed that the inhibitions ofproliferation on K562 cells were increased with the increase of concentration of anti-LA18 antibody. Eighteen the hours 3H-TdR releasing method to test the cellular toxicity of anti-LA18 antibody on K562 cells showed the activity of the anti-LA1(?) antibodies against K562 cells depended upon its dose.
