摘要
为改善肺癌化疗效果,用FCM技术分析研究胃肠外营养(PN)对人肺腺癌裸鼠模型LAX-83细胞动力学的影响。腹腔注射PN 1ml,每8小时一次,共72小时,分别于投药后24、48和72小时取佯分析细胞周期各时相细胞百分比。对照组和投药后24、48、72小时DNA合成期细胞百分比(S%)分别为43.1±3.2%、53.4±10.2%、36.4±4.8%和56.6±9.7%。S%随时间呈波浪式变化,而且48小时和72小时的S%与对照组之间有显著性差异(P<0.05)。结果提示PN可影响肺癌细胞动力学,为改善肺癌细胞对周期特异性化疗药物反应性提供契机。此外,S%呈波浪式变化根示在PN刺激下,细胞周期中的部分细胞被同步化。
In order to improve chernotherapeutic effect on lung cancer, FCM technique was employed to study tumor cytokinetics of nude mice-bearing human adenocarcinoma of the lung when administered with parehteral nutri-tion(PN) 23 animal models came into experiments. Sampling for cytokinetics analysis were performed post-administration 24h,48h and 72h. Percentages of S-phase cells (S% ) measured by FCM were 53.4 ± 10. 2(24h), 36. 4 ±4.8(48h) and 56 .6± 9.7(72h) respectively. Comparing with the control(43.1±3.2),S% at 48h decreased signif-icanfly(P< 0.05)and 5% at 72h increased significantly( P< 0.05). The results suggested that PN might regulate tumor cytokinetics and many tumor cells in cycle were synchronized. It was implicated that additional PN manipulation might increase cancer responsveness to chemotherapy.
