摘要
以苹果砧木八楞海棠(Mainsmicromalus)为材料,对体细胞无性系的再生途径进行研究。结果表明细胞分裂素BA(1.0mg/1)对茎尖的初始分化有促进作用。在丛芽的继代培养过程中应对BA浓度进行调整,以0.5mg/1为宜,BA2.0mg/1与NAA0.2mg/1的激素配比,可诱导叶柄组织分化不定芽,KT0.1mg/1,NAA2.0mg/1及2.4—DI.0mg/1的激素配比可诱导无芽茎段脱分化,BAI1.0mg/1,NAA0.2mg/1及PH7.0可诱导愈伤组织分化成不定芽。
Three different ways of somaclone formation were established in Mains micromalus. Our results showed that the best medium for induction of initial differentiation of shoots was MS +BA 1. 0mg/1+IBA 0. 2mg/l. The optimum concentration of hormorn was BA 0. 5mg/l for subculture of cluster-buds, for differentiation of leaf stalk was BA 2. 0mg/l with NAA 0. 2mg/1,whilethat for callus formation was KT 0. 1mg/1 +NAA 2. 0mg/1+ 2. 4-D 1. 0mg/1. Adventitious budsformed from calli, cultured on the MS medium added with BA 1. omg/l+NAA 2. omg/l (pH ̄ 7.0).
