摘要
本文采用高效毛细管电泳(HPCE)探索了单核苷酸的分离最佳条件:柠檬酸——柠檬酸钠缓冲液(PH3.3)为电解液,工作电压-30kv,电流20μA,毛细管长40cm,检测波长260nm,十六烷基三甲基溴化铵(CTMABr)表面活性剂为电解液添加剂。在选定条件下,分析了两个实际样品中的单核苷酸。检测限为1ng/mL。本方法具有速度快、进样量少的特点。
The optimum separation conditions of mononucleotides with HPCE was studied.Citrate buffer was used as the electrolyte solution.Cationic surfactant cetyl trimethyl ammonium bromide was applied as the electrolyte additive.Negative valtage-30KV was chosen and the electric cur- rent was 20μA.The capillary was 40cm in length and the detection was at 260nm.Two real sam- ples were analyzed with the optimum parameters.The proposed thchniqtie has the advantages of being fast and needing little amount of sample.
