添加植物抗冻蛋白提高猪胚胎冷冻保存成活率的初步试验 被引量:5

摘要 本试验采集7天猪扩张囊胚和孵化后囊胚,在1.5M甘油冷冻液中添加二种浓度(0.5mg/ml,5mg/ml)植物抗冻蛋白(AFGPs),用慢速降温方法冷冻。猪胚胎冷冻-解冻后,体外培养12小时,发育率分别为25%(1/4)和80%(4/5);24小时发育率分别为0%(0/4)和40%(2/5),孵化囊胚率分别为0%(0/5)和20%(1/5),而对照组冷冻-解冻后培养发育率12小时和24小时均为0%(0/4)。试验表明,在冷冻液中添加一定浓度的植物抗冻蛋白,可提高猪胚胎冷冻-解冻后的成活率。 In this test,dilated blastula at the 7th and blastula after hachingwere collected from swine. AFGPs with 2 concentration levels (0. 5mg/ml,5mg/ml) were added to freezing liquid of 1.5M glycerine to slowlyfreeze embryos. After freezing and thawing, the development rates were25%(1/4) and 8096(4/5) cultured for 12hrs, and 0%(0/5) and 20% (1/5) cultured for 24 hrs. The development rates of the control culturedboth for 12 and 24 hrs were 096(0/4). The test indicates that addingAFGPs to freezing liquid is able to increase the survival rate offrozen embryos.
