消化酶、固定剂对石蜡包埋肝组织DPC和FN—PAP法染色的影响

EFFECTS OF FIXATIVES AND DIGESTIVE ENXYMES ON PAP METHOD FOR DEMONSTRATING DESMIN—POSITIVE CELL AND FIBRONECTIN IN PARAFFIN SECTION OF HEPATIC TISSUE

本文作者对不同固定剂和消化酶对石蜡包埋肝组织desmin阳性细胞(DPC)和纤维联接蛋白(FN)PAP法染色的影响进行了比较研究。所试固定剂中,除Carnoy液外,其它皆需酶消化。对FN染色以缓冲福尔马林或4％多聚甲醛固定、0.2％胃蛋白酶消化2 h为宜。DPC染色以缓冲福尔马林或4％多聚甲醛固定、胃蛋白酶与胰蛋白酶双重消化各45 min最为理想。结果提示石蜡包埋组织PAP法染色显示细胞内外抗原时应注意选择适宜的固定剂和消化酶。

Compared studies on effects of fixatives and digestive enzymes on PAP method for demonstrating desmin—positive cell(DPC) and fibronectin in paraffin sect/on of hepatic tissue were done. In studied fixatives, except Carnoy fixative, the rest all needed enzyme digestion. Fibrbnectin staining with buffer formalin or 4% paraformaldehyde fixation and 2% pepsin diges- tion for 2 hours was good. DPC staining with the same fixative to fibronectin staining and double digestion with pepsin and trypsin was first chosen. The present results show that choosing the optimum enzyme digestion and fixative for staining of entercellular and extrocellular antigens with PAP method is essential.
