摘要
小苍兰脱毒苗在试管中形成球茎,可减轻组培苗出试管的工作难度和工作量:并降低了污染和重新感毒的机率;为脱毒苗的批量生产创造更好的条件。小苍兰脱毒苗的试管成球程序是:先取脱毒试管苗以培养基A(MS+BA2mg/L+NAA0.1mg/L)促使其增生许多分蘖;而后取各分蘖以培养基B(MS+BA2mg/L+NAA0.1mg/L+蔗糖60g/L)促使其基部形成小球并增大至一定大小;最后取成球苗株以培养基C使其成长与成熟。
Freesia virus-free plantlet can be obtained from meristem culture in vitro. Corms have been produced form virus-free plantlet in flask in this experiment. It reduced the ratio of contamination and infection, and decreased the difficulties and production expense when cultured plantlet are transplanted to soil. So it create a better condition for large-scale production of vius-free Freesia via tissue culture. It will be more useful after we can raise dormant corms in flask. The procedures of raising corms from Freesia virus-free plantlet in flask are as follows: 1. Using media formulation A(MS+BA 2mg/l+NAA 0.01mg/l) to tiller and increase the number of virus-free plantlet. 2. Using B(MS+BA2mg/l+NAA0.1mg/l+Sucrose 60g/l) to induce plantlet to produce corm and increase size of the corm. 3. Using C (1/2MS+NAA0.1mg/l) to induce roots of the corm.
