摘要
以松节油做生物制片的透明剂,经用石蜡切片法、徒手切片法、压片法和整体封藏法对被子植物、裸子植物、蕨类植物、苔藓植物、藻类植物和菌类植物制片试验表明,所制作的玻片标本和用二甲苯制作的一样清晰。本研究还设法使松节油与较低浓度的酒精相溶合,因而能将制片中脱水、透明两步合为一步,简化制片程序,提高工作效率。
Turpentine can dissolve paraffin, and it can clear quickly, and is volatile as well as nontoxic, so it is suitable to be a transparent agent in botanic sectioning. The sectioning materials with angiosperm, gymnosperm, pterido- phyte, pteridophyte, bryophte, algae and mushroom by means of paraffin, hand, press and whole sealing methods indicated that the slide preparations using turpentine were as transparent as those with xylene. Because turpentine can mixed up with low concentration of alcohol, both dehydration and clearing can be combined, resulting in a simplified procedure and a improved working efficiency.
