微量粘多糖快速测定法的改良及其临床应用

摘要 粘多糖可与Alcian Blue阳离子染液反应生成均匀的蓝色复合物。根据这一原理,本文采用另一型Alcian Blue染料,并选择2%醋酸作为相应的溶剂,建立改良分光光度法定量测定微量体液中粘多糖的水平。在实验过程中,染液的稳定性较原法有所提高。应用该法测定了几种正常人体液及4例MPS患儿尿粘多糖水平。本法简便,灵敏,样本用量少,可用于临床和产前诊断。 A simple modified spectrophotometric method is described for estimating microgram quantities of glycosaminoglycans (GAG). This method is based on that GAG can form complexes with Alcian Blue dye. The GAG values for normal child urine, cerebral spinal fluid, serum and amnionic fluid were presented. Total urinary GAG from MPS patients were also determined. The method is direct, rapid, sensitive, only a very small amount of sample is needed. It is suggested to be useful tool in the clinical and prenatal diagnosis of MPS.
