胰高糖素-胰岛素合用对实验性肝损伤小鼠的保护作用

PROTECTIVE EFFECT OF GLUCAGON-INSULIN ON HEPATIC INJURY INDUCED BY D-GALACTOSAMINE IN MICE

摘要 本工作采用昆明种小鼠,用D-氨基半乳糖(D-GAL)损伤肝,观察联合应用胰高糖素-胰岛素(GI)对动物的保护作用,结果如下:预先给予和不给予GI相比较,给予GI不仅降低D-GAL对小鼠的致死率(25.0%vs.48.9%,P<0.05),而且使D-GAL所引起的动物血清AST和总胆红素含量增高的幅度明显降低(225.65±69.14 ∪/L vs 497.27±193.22∪/L,P<0.01;及0.11±0.03mg%vs.0.17±0.04mg%,P<0.001),肝组织损伤程度亦明显减轻。但血清总蛋白含量则无改变。这些结果提示,GI合用可明显减轻D-GAL对小鼠肝脏损伤的程度,故可以认为GI对肝具有保护作用。 The protective effect of synergistic use of glucagon andinsulin(GI)on hepatic injury induced by D-galactosamine(D-GAL)wasinvestigated.The results were as follows:The mortality rate of themice pretreatment with GI was significantly lower than that of treatedwith D-GAL alone(25.0%)vs.48.9%,P<0.05).The AST activity andtotal bilirubin content in serum were significantly decreased in themice treated with GI as compared with those of no treatment as well(225.65±69.14 U/L vs.497.27±193.22 U/L.P<0.01;and 0.11±0.03mg% vs.0.17±0.04mg%,P<0.001).The hepatic histological finding ofthe mice were also improved,but total serum protein had no alteredafter the GI treatment.We conclude that GI therapy may reduce thehepatic injury induced by D-GAL,so that the GI might exert protectiveaction on liver.
