摘要
本文以电镜及细胞化学Ca^(2+)显示法,结合电子探针显微分析,观察到654-2(山莨菪碱的人工合成品)在减轻离体大鼠心钙反常心肌细胞超微结构损伤的同时,使大部分细胞膜结合Ca^(2+)保存及线粒体内Ca^(2+)积聚明显减轻。结果提示,654-2对Ca^(2+)反常心肌细胞的保护作用与阻止细胞内Ca^(2+)积聚及异常分布有关。
A lot of evidences showed that anisodamine (A) hasa protective effect on ischemic and anoxic myocardial injury. But the mechanism remains unclear. This study was to observe the effect of A on calcium paradox (CP) and intracellular calcium distribution in isolated rat hearts. The CP model in isolated rat heart was established in accordance with the method of Van Gilst, et al(1987), Animals were(?)andomly divided into 3 groups, (1):control(n=6),(2):CP (n=6).(3): CP+A(n=6). The following parameters were studied: (a) ultrastructural changes: (b): cytochemical localization of calcium (with phosphate-pyroantimonate method); (C): elements microanalosis (by electron probe). It was found that the ultrastructural damages of cardiac myocytes induced by CP were markedly decrensed, sarcolemma-bound calcium in the majority of myocytes was preserved and calcium accumulation in the mitochondria was decreased when the perfusing solution was supplemented with A at the periods of stabilization and low calcium.
The results suggest that the protective effect of A on CP of isolated rat heart is closely related to the prevention of intracellular accumulation and abnormal distribution of calcium and sarcolemma-bound calcium plays o role in the maintenance of the integrity ofmembrane and ultrastructure of cardiac myocytes.
