摘要
本文报道了培养物中脱氧雪腐镰刀菌烯醇的提取、纯化的方法及其结构鉴定.大米培养物用水润湿,再用氯仿:甲醇(8∶2)浸泡或80%乙醇渗滤,得粗提物.经一次硅胶柱层析后,用醋酸乙酯-正己烷结晶,得白色结晶.根据它的熔点(151~155℃)、分子量(296)、EI质谱、~1H核磁共振光谱及红外光谱,确证结晶为脱氧雪腐镰刀菌烯醇.气相色谱测定其纯度,含量为98%.
Method of extraction,purification and structural identification of deoxynivalenol (DON) has been reported.Rice culture was first moistened with water,and then soaked in chloroform-ethanol (8∶2)or percolated with 80% ethanol.Crude toxin was extracted from the culture.White crystalline toxin was obtained from crude toxin by silica gel column chromatography and crystallization with ethyl acetate-n-hexane.According to its m.p.(151-155℃),MS,~1HNMR and IR. the crystalline toxin was ascertained as deoxynivalenol,Its purity was determined by gas-liquid chromatography to be 98%.
