大鼠颌下腺内神经元特异性烯醇酶阳性细胞的分化

摘要 用神经元特异性烯醇酶(NSE)免疫组织化学染色法,观察生后各发育阶段大鼠的颌下腺,发现NSE阳性反应主要定位于颌下腺的纹状管和颗粒曲管(GCT)细胞,染色强度随年龄增长而加强。电镜观察表明NSE阳性反应在多颗粒细胞最强;少颗粒细胞次之;无颗粒细胞极弱或阴性。NSE阳性反应产物主要分布在分泌颗粒之间的胞质中,以及线粒体和内质网膜的表面,细胞核和分泌颗粒以及其他细胞器均为阴性。显微分光光度计测定显示纹状管与GCT中NSE相对含量从生后2周的1.61±0.11增加到成年时的5.95±0.81。各年龄组间酶含量的差异有显著的统计学意义(p<0.01)。 The postnatal development of the rat submandibular gland was studied by means of the NSE immunohistoohemioal method. We found that the positive reaction occurred mainly in cells of the striated ducts (SD) and granular convoluted tubules (GOT). With increasing age, the intensity of positivity became stronger and stronger. Electron microscopy, showed that polygranular cells were intensely in positive, oligogranular cells wero moderately positive, while the non-granular cells reacted very weakly, were negative. Positive NSE deposits were mainly located in oytosol around the gee retory granules as well as on tho surface of mitochondria and endo-plasmio reticula. No positive reaction could be observed within the nucleus or other cell organelles, including seeratory granules. Microspeotrophotometrio measurmenbs showed that NSE activity of SD and GCT cells increased from 1.61 at 2 weeks to 6.95 in adults. The differences in enzyme activity between each age group were statistically significant.
