正常小鼠血清中三种免疫球蛋白的检测

摘要 实验小鼠已成为免疫学研究的有力工具,不同品系个鼠在发生肿瘤和自身免疫病方面,有其品系的特征,据推测这种疾病的遗传与免疫球蛋白的合成量有关。而小鼠免疫球蛋白的含量随品系而异。因此在分析体液免疫反应时,首先要考虑小鼠品系的免疫球蛋白正常水平。迄今有关不同品系小鼠免疫球蛋白的遗传背景和因增龄而引起变动的资料为数不多,其主要原因是各类免疫球蛋白的抗血清制造较难,供应不多。本文目的是用自制的抗小鼠IgG(Fcr)、IgA、lgM血清,通过琼脂单向免疫扩散和速率散射比浊法,检测普通级小鼠中3类(IgG、IgA、IgM)免疫球蛋白,以了解其在不同品系。 This paper reports the level of serum Igs(G, A, M) in coventional NIH, BALB/c mice of different ages and sex detected by routine radialimmunodiffusion (SRID)and rate scatted nephelometry. The level of serum Igs (G. A. M) of mice increased with age. The content ofIgG was 10.73 mg/ml, IgA 0.41 mg/ml, IgM 0.82 mg/ml in NIH mice and IgG7.74 mg/ml, IgA 0.34 mg/ml, IgM 1.35 mg/ml in BALB/c mice at the age oftwo months, then the level continued to rise steadily. After 6 months IgG of NIHmice and IgM of BALB/c mice continuely increased, the others tended to decline.Except IgM there was a significant difference between sex at 6-12 months of age. nosignificant difference in the other ages of two strain of mice. The results obtained from SRID and nephelometry were corralated well (IgG r=0.931, IgA r=0.861). The level of serum Igs (G. A. M) among 5 different strainsof mice at two month ages was detected by rate scatted nephelametry. In differentstrains. no significant difference was noted in IgA level. The level of IgG in BALB/c mice was the lowest and the level in kunming was the highest. The IgG levelranged from 9.16-10.73 mg/ml in the other 3 strains. In addition, IgM of DBA/2mice show a significant difference bdtween sex among 5 strains. (t=3.5051 p<0.05). The present study established Igs criteria of mice in body fluid immune functionand provided experimental reference data of mouse body fluid responce in experime-ntal immunology and pharmacological immunology.
