Cryopreservation of Human Fetal Liver Cells and Clinical Application

102例人胎肝细胞在液氮中(—196℃)保存1—1257天后,平均细胞回收率、CFU—GM回收率和台盼兰拒染率分别为98％、71％和62％。18例再生障碍性贫血患者输注了冻存胎肝细胞,9例显示了较好的治疗效果;2例白血病患者冻存胎肝移植成功;冻存胎肝细胞输注对改善肝脏疾病的临床症状有一定好处。

102 cases of human fetal liver cells were stored in liquid nitrogen for a period of 1—1257 days. The viability of cells after thawing as evaluated by means of total nucleated cells recovered (98%), trypan blue exclusion rate(62%) and CFU-GM recovered (71%) shows that cryopreservation technique is an effective measure to preserve the cells concerned. So far 26 patients suffering from aplastic anemia, leukemia or liver disease have receivd the infusions of cryopreserved fetal liver cells and resulted beneficial effects without causing harmful side reactions. A temporary engraftment as evidenced by the presence of donor type karyotype in two children with leukemia received cryoprescrved fetal liver cells transplantation was observed.
