钴—60辐照增加金黄地鼠对EHFV敏感性的研究

摘要 流行性出血热的发病机制至今尚未得到阐明,其关键是没有较理想的动物模型。本实验的目的在于研究建立EHF实验动物模型和选择适宜的人工干预方法,以增加其敏感性,为发病体征观察打下基础。实验证明,我们选择^(60)Co辐照金黄地鼠能大大地提高其对EHFV的敏感性,脏器中抗原检出率非常明显地高于未照射动物(x^2=8.624,P<0.01)。通过不同照射剂量的比较,以200Rad/只最为合适,既可全部检出定位抗原,又不影响抗体产生,且无非感染死亡。 Up to now, the pathogenesis of EHF has remained unknown. The main reason for this is that there are no ideal experimental animal models. The purpose of our experiment is to find out a suitable method of man-made condition to increase the sensitivity of the experimental animals to EHFV. The result of our experiment shows that radiating the Golden hamsler with ^(60)Co can greatly increase its sensitivity to EHFV. The detected rate of EHF antigen in the viscerals of the radiated Golden hamslers (24/24) is obviously much higher than that in those un-radiated(8/14) (X=8.62, p0.01). Comparing the results of the different dose of radiation, we found that 200rad/pen Golden hamsler is the most suitable. At this dose, all the location antigens can be found, and meanwhile the yeild of the antibodies to EHFV can't be effected. The titers of antibody to EHFV in group of radia tion 200rad, group of radiaticn 400rad and, group of injection are 1:320-1:640, 0-1:160 and 0-1:160 respectively. And what is more, there is no non-infected death occured. This indicates that the method of radiating the Golden hamslers is much better than that of injecting cyclophosphamide into Golden hamsler. It laid the foundation to have further observation of the clniical manifestation of the infected animals.
