摘要
本文用 DNA 斑点杂交法和 Southen 印迹法对脑瘤中 erbB、sis、myc 和 fos 这四种癌基因的扩增和重排进行了研究。发现 erbB 基因和 fos 基因在恶性胶质瘤中扩增显著(>50倍)。脑膜瘤和脑血肿组织中,这四种癌基因扩增都很显著,而少枝胶质瘤中这四种癌基因的拷贝数均较少。另外还发现脑瘤中这四种癌基因均有重排,并呈现一些对脑瘤比较特异的杂交片段。本文还对肿瘤的发生机制、癌基因扩增和恶性程度之间的关系等进行了探讨。
Rearrangement and amplificarion of erbB,sis,myc and fos in brain tumors are studied with DNA dot blot and Southern blot analysis.Apparent amplification(>50 folds)of erbB and fos genes in malignant gliomas and of these four oncogenes in meningiomas and brain hematoma tissues is found.However fewer copies of these four oncogenes in oligodendrogliomas are found.Rearrangement of these four oncogenes in brain tumors and some hybridized-fragments more specific for brain tumors is also found.The mechanism of carcinogenesis,relationship between amplification of onco- gene and malignant degree are discussed.
